Does serial determination of lactate dehydrogenase isoenzymes 1 and 2 ratios contribute to the diagnosis of acute myocardial infarction?

A commercial kit for the manual quantification of slight albuminuria by immunoturbidimetry (“AlbuSure-Qnt”; Cambridge Life Sciences, Cambridge, U.K.) has been adapted for use on the “Multistat 3 Plus” centrifugal analyzer (Instrumentation Laboratory). Standards with concentrations of 0, 10.3, 19.0, 40.0, 79.0, and 165 mgfL are supplied ready for use. Antiserum is diluted with buffer in the ratio 1:6.8 to provide a single reagent. The sample/reagent volume ratio is the same for both manual and automated procedures. For the centrifugal analyzer method, 50 p.L of undiluted antiserum is sufficient for one rotor (20 cuvettes), substantially increasing the number of tests per kit. The incubation time is decreased from 30 mm to 4 mm. Immune-complex formation was initially monitored at 405 nm according to the manual procedure, but the absorbance change was found to be greater at 340 nm, the wavelength subsequently used.
